Pharmacodynamics/Kinetics:
Bioavailability: increases with chronic dosing

Half-Life Elimination: 40 hours

Excretion: Urine

Absorption: In a single-dose pharmacokinetic study, Ribavirin taken with a high-fat meal (841 calories, 53.8 grams (g) fat, 31.6 g protein, and 57.4 g carbohydrate) increased drug absorption. Coadministration of Ribavirin with an antacid containing magnesium, aluminum, and simethicone resulted in a decrease in drug absorption. The primary toxicity of Ribavirin is hemolytic anemia.

Dosage Forms: Powder for injection: 50mcg/0.5mL, 80mcg/0.5mL, 120mcg/0.5mL, 150mcg/0.5mL.
 
Usual Dosage: <45kg: 40mcg; 46-56kg: 50mcg; 57-72kg: 64mcg; 73-88kg: 80mcg; 89-106kg: 96mcg; 107-136kg: 120mcg; 137-160kg: 150mcg.

Administration: For subcutaneous administration; rotate injection site.

Monitoring Parameters: Baseline and periodic thyroid stimulating hormone, hematology (including CBC with differential, platelets), and chemistry (including liver function labs) testing.

Precautions: Users should drink lots of fluids during initial treatment to reduce nausea and dehydration. Diabetes, hyperglycemia, and decreased platelets have been observed. Safety and effectiveness in HIV-positive people, in pregnant women and pediatric patients is not adequately established.
